Spawn an eye at Eris Morn, then head to Hellmouth, on the other side of the area. Head to the Anchor of Light and continue straight on following the trail to Hellmouth. Get off your sparrow and go down into the Hellmouth. Take a left to go to the Gatehouse. Continue to follow the passage and head to the illuminated door to the right in the next area. This passage brings you into a room with enemies.
Run to the back of the room and prepare for a fight. An enemy called The Eternal Blade will collapse in the room. It hits hard and has a significant health bar, so you want to be pretty close to 900 Potency before you take it. When you kill him, you get the Bound Manacle item you need.
The other part of this essence is getting 100 Kills with Grenade Launchers in Gambit. This task should not take too long. It took me two laps to finish. Just watch the moment the heavy ammo spawns and grab it as soon as it spawns. Then, gather groups of enemies behind you and take as many as you can with a grenade launcher.
